I would go by car, with a GPS, and allow yourself the whim of making a left or a right to explore. The GPS will correct your path anyway. I prefer Verona to Venice, Sienna to Florence, and something like Perugia to Rome simply because it avoids the tourist crowds.

But if you simply want to go sightseeing, and see the major sites, I would consider a guided tour bus and just go along with the tourist crowds. You wouldn't have to worry about driving a car, and they'll take care of your schedule and itinerary for you.
